Abstract

Lesions and diseases of the musculoskeletal system represent more than 6 million consultations in medical emergencies in US hospitals. The damage and repair process is similar in most cases, but the functional recovery of a muscle injury varies according to the type of the trauma and the most serious injuries can progress with the formation of fibrous tissue interfering with adequate muscle contraction and it can lead to muscle and chronic contractures. Current treatment has a low efficacy for the prevention or treatment of complications such as atrophy, contracture and pain followed by functional loss and post-traumatic fibrosis. One of the main fibrogenic factors is TGFb1, which is the main agent for the formation of pathological fibrosis. The expression of factors that inhibit TGFb1 is very relevant. Thus, the present project proposes the construction of a lentivector expressing the soluble TGFb1 type II receptor (sTbRII) under the control of the NF-kB-inducible promoter and standardization of the system in 2D and 3D muscle cell primary culture.
