Abstract

The objective of this project will be conduct dose titration studies with Plexomin L-Mn to determine the effects of added Mn and lysine on satellite cell GLUT- 4 concentrations, differentiation status and myosin heavy chain expression (MHC I). In this project, the semimembranosus from 2 steers with 16 months, will be used to isolate bovine primary satellite cell (BSC). Myogenic differentiation will be induced using 3% horse serum upon cultured BSC with increasing doses (0, 0.1 ¼M, 1.0 ¼M, 10 ¼M, or 100 ¼M) of Mn, and the treatments will be done in triplicate. After 96 h of incubation, cells will be harvested and used to measure the gene expression of AMPK±, ²1-AR, ²2-AR, IGF-1, MHC I, MHC IIa, and MHC IIx, glucose transporter 4 (GLUT4) and relative to ribosomal protein subunit 9 (RPS-9).
