Abstract

Prostatic carcinoma is aggressive glandular neoplasia, highly metastatic in humans and canines. Usually diagnosed in dogs between 8 and 11 years old, more frequent in whole males, but can occur in neutered males. As it presents similarities in histological aspects with human prostate carcinoma and is the only species, besides man, that can occur spontaneously, the dog is a good model of comparative oncopathology for genetic, epigenetic, and therapeutic studies. Since current therapeutic protocols are based on delaying the progression or performing surgical excision of the tumor mass, it is not possible to obtain a favorable prognosis, new alternatives should be investigated, especially those that may present lesser side effects to the animal, less environmental contamination and greater safety for tutors. The latex of H. drasticus is used by the vast majority of the population through popular information for curative, preventive and/or palliative purposes. In this context, the essential oil of Himatanthus drasticus (adc) woodson, presents natural compounds that have the potential to exhibit important cytotoxic activity and in the culture of carcinomas (in vitro). This project has a partnership with the Federal University of Alagoas, UFAL, with Professor Thiago Barros Correia da Silva, who already has the essential oils characterized. In this study, two different cultures originating from primary canine prostatic carcinoma will be used, in a standardized two-dimensional primary culture, from previous projects carried out by our research group (Kobayashi et al., 2020), as well as a human prostatic carcinoma lineage (PC3), to test the cytotoxic effect of the essential oil of Himatanthus drasticus (adc) woodson. The experimental groups will be composed of a control group (untreated) and a group treated with the essential oil of H. drasticus in different concentrations, to reach the IC50. Subsequently, the synergistic effect of essential oil will be tested in cultures treated with conventional chemotherapy (Doxorubicin and COX-2 inhibitor). The objective of this work is to test the antitumor potential of H. drasticus oil in vitro cultures of canine and human prostatic carcinoma (PC3). Cell proliferation and migration tests will be performed, in addition to the Invasion Potential of the cell by the Transwell Test test. (AU)
