Role evaluation of miR-199a and miR-148a in cell migration in Thyroid Cancer

Abstract

Thyroid Cancer (TC) due to several genetic changes confer a proliferative advantage to the follicular cell. Platforms based on these changes have been improving the diagnosis, however, do not focus on prognosis analysis and routine assessment of the possibility of cancer recurrence and metastasis. The expression of microRNAs (miRNAs) on the routine evaluations can help this issue, given the importance of these small RNAs in the development and evolution of several cancers including TC. In a project of my own and oriented by Prof. Dr. Laura Sterian named "In silico analysis of miRNAs Related to thyroid carcinogenesis", still in progress, presented as partial results hsa-miR-199a (both strands 3 'and 5') and hsa-miR-148a-5p as down-regulated miRNAs in papillary carcinomas (PTC) with lymph nodes metastasis (LN) and medullary thyroid carcinomas (MTC) LN samples in comparison with respective normal thyroid tissue. These findings corroborate previous studies evaluating the relationship between invasion and metastasis. In the project proposed, we aim to evaluate the role of these miRNAs in thyroid cell migration and invasion using two TC cell lines (TPC-1 and BCPAP) by the monolayer and Boyden Chamber lesion assays. The statistical analysis and construction of the graphs will be performed in software GraphPad Prism version 9.0 software. (AU)
