The role of microRNAs in the regulation of C1orf24 expression in thyroid tumors

Abstract

Using SAGE (Serial Analysis of Gene Expression), we have previously showed that C1orf24 (Chromosome1 Open Reading Frame 24) is highly expressed in thyroid carcinomas compared to benign lesions. C1orf24 was able to distinguish benign from malignant thyroid lesions with high sensitivity (97%) and specificity (90%), which suggest its application as pre-operatory diagnostic markers. Although it may have relevancy as biomarker, the molecular mechanism underlying C1orf24 expression remains unclear. One of the post-transcriptional mechanisms of gene regulation is the action of the microRNA (miR). miR expression varies according tissue, development stage and cancer types. Therefore, using three different databases, our aim is to identify miRs that potentially regulate C1orf24 expression. We next, will further validate the expression of miR in thyroid lesions and correlate with C1orf24 expression. To investigate whether the selected miR modulate the C1orf24 expression, they will be transiently transfected into a thyroid carcinoma cell line that show to have high levels of C1orf24 expression and low level of the selected miR. Additionally, to confirm the direct effect of miR in the C1orf24 expression, we will introduce mutations into 3'-UTR do mRNA of C1orf24 and test whether or not the miR is able to modulate C1orf24 expression. Finally, we will test whether the miR re-expression changes the cell growth and apoptosis rate and modulate the expression of gene potentially regulated by C1orf24. (AU)
