Impact of feed supplementation with tangeritin on lipopolysaccharide-induced inflammation in mice periodontal tissues

Abstract

Drugs that modulate the immune response are constantly being investigated in the treatment and prevention of immune-inflammatory diseases, however, they can have a variety of disadvantages, including side effects and low efficacy. Natural products such as flavonoids have gained the interest of researchers due to their beneficial pharmacological and therapeutic activities. This project aims to evaluate the hypothesis that feeding supplementation with tangerine (TAN), a citrus flavonoid that has a potential antioxidant and anti-inflammatory effect, could attenuate inflammation and consequent alveolar bone loss in an animal model of periodontal disease induced by lipopolysaccharide (LPS) injections. For this, 50 mice (BALB/c) will receive a daily conventional diet or diet enriched with TAN (50 and 100mg/kg). A positive control group will receive a diet enriched with naproxen (NSAID, 10mg/kg). After 30 days of feed supplementation, the animals will be submitted to periodontal inflammatory disease induced by micro-injections of LPS in the gingival tissue adjacent to the palatal surface of the molars, 3 times a week for 28 days. After the experimental period, the animals will be euthanized and the jaws collected for stereometric analysis; evaluation of pro-and anti-inflammatory cytokine expression by enzyme-linked immunosorbent assay (ELISA); measurement of oxidative damage (MDA), and quantification of bone loss by µCT. At the end of the study, it is expected to propose strategies capable of collaborating in the prevention and treatment of diseases that affect the periodontal, favoring the maintenance of periodontal health.
