Abstract

Currently, milk pricing is based on milk volume and quality. Milk fat is one of the most important milk components considered in the establishment of the final milk price. Consequently, researchers intend to better understand the effect of nutrition on the milk fat synthesis in the mammary gland. Recent studies showed that amino acid supplementation in diets of dairy cows affects milk fat yield and this effect have been related to the role that amino acids play in regulating the action of the mammalian target of rapamycin complex 1 (mTORC1), which is related to sterol regulatory-element-binding protein 1 (SREBP-1), an important transcription factor involved in regulating de novo fatty acid synthesis in mammary gland cells. However, this hypothesis linking amino acids and SREBP-1 has not always been confirmed, which in turn may be related to the interaction among amino acids, as well as amino acid supplementation levels. Thus, the objective of this study is to evaluate the effects and doses of essential amino acids in de novo milk fat synthesis in bovine mammary epithelial cells, at physiological levels. The study will use isotopically labeled acetate (13C-acetato) as a tracer. In this experiment, we will evaluate the individual effect of essential amino acids in de novo fatty acid synthesis in bovine mammary epithelial cells subjected to 12 treatments varying the amino acid profile. Understanding the link between amino acids and milk fat synthesis in the mammary gland has practical application in the diet formulation of dairy cows.(AU)
