Abstract

Immunoglobulin G (IgG) is a glycoprotein also identified as an antibody, being used in several therapeutic indications, for that it must have high concentrations and purity for certain processes, this implies in some techniques aimed at selective adsorption that is recommended for its process of purification. This project aims to evaluate the interaction of human IgG in fibroin microparticles (adsorbent), which is a natural polymer that has not yet been studied in the literature. The fibroin microparticles will be prepared by the atomization method in an ethanol coagulation bath and characterized in terms of morphology, particle size, and thermal and chemical properties. The adsorption will be carried out at room temperature under different conditions. Biological buffers will be used varying the pH between 5.0 and 9.0, all at 25 mmol L-1 in the experiments. The modeling of the adsorption isotherms will be carried out through the classical nonlinear models of Langmuir, Langmuir-Freundlich, and Temkin, which will be compared and verified with the model that best describes the adsorption. Finally, the dynamic binding capacity will be determined by breakthrough curves. Thus, with the results obtained, it will be possible to study new cases of application of fibroin microparticles and improve Immunoglobulin G purification techniques.(AU)
