Effects of prebiotic therapy with polydextrose prebiotic on experimental periodontitis: study of histological and immunohistochemical profile in rats

Abstract

Considering the ecological plaque hypothesis, periodontitis will inevitably recur, unlesspredisposing factors that lead to changes in the microbiome are treated. A potential approach to promote oral health is to restore homeostasis of the resident oral microbiota and modulate the host's immune response. Prebiotics (PREBs) stimulate the growth and metabolic activity of beneficial microorganisms beneficial to health, promoting resistance to dysbiosis, and restoring the state of health. Prebiotic therapy represents an interesting therapeutic approach as an adjunct to standard periodontal therapy. Knowledge about the possible effects of PREBs on oral health is recent and needs further studies in Dentistry. The purpose of this study will be to evaluate the effects of polydextrose prebiotics on the development and progression of experimental periodontitis(EP) in rats. 44 male Wistar rats will be used, randomly divided into 4 groups (n = 11):1) control (C) - animals without EP and that will not receive prebiotic (PREB)administration; 2) EP - animals with EP and that will not receive PREB administration;3) PREB - animals without EP and that will receive PREB administration; and 4)EP/PREB - animals EP and that will receive PREB administration. From day -30 until the end of the experiment, the prebiotic polydextrose will be added daily to the drinking water for the animals of PREB and EP-PREB groups, until the end of the experiment. On day 0, EP will be inducted on right first mandibular molars of the animals of EP andEP-PREB groups. All animals will be euthanized on day +14 of the experiment. Histological (hard and soft tissues profile) and immunohistochemical (TRAP, TNF-±,IL-1², IL-10, TGF-²1 e CINC) analysis of the mandible will be performed.
