Evaluation of the bone response of an Odanacatib coated implant surface in ovariectomized rats: in vivo experimental study

Abstract

The active principle Odanacatib (MK-0822) is a highly selective and sensitive inhibitor of capetpsin K (CatKI) that, currently, has been positively highlighted in the osseointegration process, due to its anti-resorptive action, which encouraged this project to evaluate the response implant surface bone coated by double acid etching and Odanacatib (MK-0822) in ovariectomized rats. Thus, in this study, 32 rats (Albinus, Wistar) will be used, divided equally into two groups: ovariectomized (OVX) and SHAM (placebo). Each animal will receive two implants, one in each tibia, the total number of implants to be used is 64 implants, having their surfaces treated by double acid etching and Odanacatib at 0.06 mg/ml, through the biomimetic method, thus forming 4 groups: double acid attack in SHAM rats (CST), double acid attack in OVX rats (COT), MK-0822 0.06 mg/ml in SHAM rats (M6ST) and MK-0822 0.06 mg/ml in OVX rats (M6OT), being 15 and 40 days the periods of euthanasia. Scanning electron microscopy (SEM) and energy dispersive x-ray (EDS) analyses will be performed after surface treatment of the implants, for topography and chemical composition analysis. At 15 and 40 days, after implant placement, histological, histometric, and immunohistochemical analyzes will be performed and quantitative data will be evaluated adopting the significance level p<0.05.(AU)
