Abstract

Cancer is a major cause of morbidity and mortality worldwide. For the beginning and progression of Cancer, collaboration between tumor cells and the tumor microenvironment is necessary, being one of the forms of communication through Extracellular Vesicles (EVs). The tumor microenvironment is composed of different subsets of cells, including neutrophils. Neutrophils are cells of the immune system, their main functions are the recognition and binding of pathogenic structures, phagocytosis, degranulation, formation of extracellular neutrophil traps, are often associated with inflammation and the fight against infections. Neutrophils are no longer considered only as cells of immediate response, as they have different functions in all stages of Cancer, both pro-and anti-tumor. These cells accumulate in the peripheral blood of Cancer patients, especially in advanced stages. Tumor-Associated Neutrophils (TANs) have been studied for their role in tumor development through the secretion of cytokines and growth factors. In addition, they can influence the formation of metastases and the suppression of antitumor immune responses. However, this role of neutrophils during the formation and progression of Cancer has not been well defined, making it the target of current research. Therefore, we propose to evaluate the effects of EVs derived from primary fibroblasts and keratinotics and biopsy of Oral Cancer in co-culture with neutrophils, investigating the potential effects regarding differentiation and activity of neutrophils. This project is expected to understand the initiation of Oral Cancer through the response of neutrophils to stimuli of vesicles treated with alcohol and, with this, explore the signaling pathways that allowed the escape of neutrophils in the face of alcohol stimuli.
