Abstract

The present proposal aims to identify protein and/or enzymatic selenium biomarkers in the species Psectrogaster amazonica (branquinha, non-carnivorous species) and Raphiodon vulpinus (curvina, carnivorous species) from the Jirau region, Madeira River/Rondônia. To achieve the proposed objectives, metalloproteomic strategies (2D PAGE - GFAAS - LC-MS/MS), Shotgun - LC-MS/MS and Bioinformatics will be used to: - Fractionate the proteome from muscle and liver tissue samples of the species Psectrogaster amazonica and Raphiodon vulpinus; Selenium map in protein spots of 2D PAGE gels; To characterize the selenium-associated protein spots in the muscle and liver proteome; Identify proteins associated with selenium and differentially expressed by Shotgun - LC-MS/MS. Based on the results of characterization of metal binding proteins and expression analysis of these, it is expected to elect possible selenium biomarkers in the species Psectrogaster amazonica and Raphiodon vulpinus from the Amazon region.
