Abstract Currently, there is an intense search for biocompatible materials that are osteoconductive, osteointegrative, with good biomechanical strength and low modulus of elasticity to be safely used as orthopedic implants. Therefore, the objective is to evaluate surface modification treatments and effects on the antimicrobial activity of Ti-34Nb-6Sn/Mg ¿ alloys produced by powder metallurgy for use as biomaterial. Triplicates distributed into seven groups will be analyzed at three different times (24h, after 3 days and 7 days), characterized by control group (GC) and 6 more distinct treatment groups containing only osteogenic medium, evaluation with surface modifications by anodization with glycerol electrolyte + 0.25% NH4 F at 30V for 9h and annealed at 530ºC (5ºC/min) and immersion coating in 0.3M CaCl2 and 0.5M Na2HPO4 solutions for hydroxyapatite growth, both in pure titanium and on the surface of the Ti-34Nb-6Sn/Mg alloy. Subsequently, both biological characterization will be performed through adhesion tests, cell viability, sample extraction for RT-PCR for gene expression, gelatinolytic activity of metalloproteinases, Alizarin Red assay and Wound Healing assay; substrate characterization by scanning electron microscopy (SEM), X-ray diffractometer (XRD) and dynamic contact angle; energy dispersive X-ray spectroscopy (EDS), inductively coupled plasma mass spectrometry (ICP-MS) analysis, and uniaxial compression tests. Antimicrobial activity will be determined by performing antibiogram, Minimum Inhibitory Concentrations (MIC/MIC) and Minimum Bactericidal Concentration (MBC/MBC), in addition to antibiofilm activity. Data normality will be assessed by the Kolmogorov-Smirnov test (p < 0.05). If there is a normal distribution, analysis of variance will be performed to compare the groups within each evaluation time point and the One Way Repeated Measures test will be performed for each group (p < 0.05). Additionally, GraphPad Prism 8 and differences between groups will be assessed using a two-tailed Student's t-test. | |
