Abstract

Colorectal cancer (CCR) is the third most common type of cancer in the world, and in Brazil, it is the second most incident and despite early screening and new treatment strategies, it remains the second most fatal cancer in the world. The intestinal microbiota has been implicated in the development of colorectal cancer. The work of our group has identified the pathogenic role of Fusobacterium nucleatum in the prognosis and, more recently, in the early diagnosis of these tumors. Thus, this project aims to assess the molecular and clinical impact of the intestinal microbiome and its role as biomarkers for early detection of colorectal cancer and its precursor lesions using for the first time MinION technology and residual stool samples preserved in a Fecal Occult Blood Analysis (FIT). Fecal samples from participants of the screening program who had positive fecal occult blood test and who underwent colonoscopy will be evaluated, with 100 participants without lesions and 100 with precursor lesions or cancer. Bacterial DNA isolated from stool samples shall be used for amplification of variable regions of the 16S ribosomal RNA gene and the libraries will be sequenced using the nanopore long-read sequencer MinION. The sequenced amplicons will be analyzed with the cloud-based EPI2ME algorithm. We intend to identify a microbiota profile associated with early diagnosis of CRC in the Brazilian population. (AU)
