Abstract

The objective of the present study is to identify and quantify the active microbial populations, their metabolic pathways and interactions within the microbiome, as well as with the host, through shotgun metagenome and metaproteome approaches on rumen and cecum digesta of Nellore cattle supplemented with narasin. Moreover, it will be evaluated the main site of action of Narasin (rumen and/or cecum), as well as if alterations in metagenome and metaproteome may be correlated with ruminal and cecal acidosis. A total of 64 non-castrated Nellore cattle will be used. The finishing diets will contain 87% concentrate and the treatments will be: 0 and 20 ppm of Narasin (Zimprova, Elanco Animal Health®, Brazil). The experimental design will be in completely randomized blocks. The animals will be slaughtered in a commercial slaughterhouse after 112 days of confinement. Samples of rumen and cecal content will be collected and destined for shotgun metagenome and metaproteome approach. Samples of ruminal and cecal epithelia will also be analyzed for morphometry and histology. An integrated metaproteome and metagenome analysis will be performed, and correlations between different proteome and bacterial communities will be assessed. (AU)
