SYNTHESIS OF POLYCAPROLACTAN (PCL) FIBER MATS INCORPORATED WITH 45S5, 58S BIOGLIDES: COMPARATIVE ANALYSIS ON OSTEOGENESIS IN VITRO

Abstract

Currently, studies have been conducted to improve the mechanical performance of bioactive glasses through their incorporation into matrices similar to the extracellular matrix, such as polymeric sheets. Thus, the high bioactivity and functional characteristics of the bio-glasses, such as bone adhesion capacity, are added to the structural characteristics of the polymers, resulting in an improvement in the mechanical performance of this biomaterial composite. Thus, the aim of this study is to evaluate in vitro the influence of ultra-thin polycaprolactone (PCL) fiber mats incorporated with 45S5 or 58S bioglides, both chlorinated and produced by the sol-gel route using different processing methods. The blankets will be obtained by the electro spinning method from a polycaprolactone (PCL) polymer solution incorporated with 45S5 and 58S biohydrogens produced by three different processing methods (freeze-drying, freeze-drying plus calcination, and calcination). For the morphological characterizations of the materials, scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS) will be used. After characterization, for the in vitro study mesenchymal cells will be obtained from the bone marrow of femurs and differentiated into osteoblasts for evaluation of the biomaterial influence on cell activity. After cell plating, analysis of cell morphology on the biomaterial, analysis of cell viability, total protein content, alkaline phosphatase (ALP) activity, and mineralization nodule formation will be performed. The data will be statistically analyzed, initially by means of the normality test in order to apply the appropriate statistical test. The level of significance adopted will be the conventional value of 5%. With the development of this project, it is expected to select the best production route for 45S5 and 58S chlorinated bioglasses to be incorporated into polymeric membranes that can be used as bone regenerating biomaterials.
