Abstract

An ideal suture thread should present characteristics such as: security in the knot, tensile strength, easy handling, low cost, minimal tissue reaction and adequate strength during the healing and tissue repair phase. Thus, studies related to the production of suture threads with such properties are increasingly frequent in tissue engineering. The objective of this study will be to produce and evaluate the suture threads produced from the PLLA/GelMA hydrogel coated with chitosan/Melaleuca Alternifolia in cellular activity in vitro. After producing the samples using the rotospinning technique and subsequent coating, the wires will be characterized by testing the surface topography of the samples, qualitative analysis of the chemical composition of chemical elements (EDS), analysis of wettability using the contact angle technique and swelling test and degradation test in saline phosphate buffer (pH 7.4) of samples prior to in vitro tests. Subsequently, the samples will be submitted to in vitro biological tests using human fibroblast cell lineage (HFF1) that will be plated with the samples in order to evaluate the influence of biomaterials on the cell activity of fibroblasts, using tests of cell morphology analysis by SEM, determination of cell viability (MTT), analysis of total protein concentration and gene expression by quantitative real-time PCR analysis. All biological assays will be performed in three independent experiments. The data will be statistically analyzed using the Graph Pad Prism software, initially using the normality test in order to apply the appropriate statistical test. The significance level adopted will be the conventional value of 5%.
