Proteomic profile of rat kidneys submitted to maternal protein restriction: Possible implications on renal morphophysiology and aging

Abstract

Pregnancy and lactation are extremely important for both the mother and the children, andseveral problems during these periods can harm the development of the offspring,affecting multiple organs and systems, including the urinary and kidneys. Such a condition isassociated with the Developmental Origins of Health and Disease (DOHaD), in which one of themodels studied is maternal protein restriction (MPR), which generates hypertension andheart problems, effects on the central nervous system and satiety, as well aschanges in metabolism. Therefore, the objective of this project is to evaluate the effects of MPRon the renal morphophysiology of rats on the postnatal day (PND) 540, from the profileproteomic. For this, male rats of the Sprague Dawley strain were dividedinto 2 groups: Rats born to mothers fed a normal protein diet (CTR, 17%protein) or hypoprotein diet (GLLP, 6%), during pregnancy and lactation, which laterconsumed normal protein diet. In PND 540, the animals were euthanized and the kidneyscollected. The kidneys will be used for morphological, morphometric,histopathology and proteomics, in order to evaluate the differentially expressed proteinsbetween the groups, integrating this profile with the morphological data. The expected resultsare that MPR directly affects renal morphophysiology with aging, which canlead to important systemic alterations, allowing susceptibility to hypertension, as well asas negative effects on other organs.
