Abstract

During the generation of recombinant monoclonal antibodies, one of the essential steps is the expression of light and heavy chain sequences in mammalian cells. The transient transfection process is usually done by co-transfecting two vectors, one cloned with the heavy chain and the other cloned with the light chain. The co-transfection is satisfactory when the balancing of the two vectors allows the expression of the two chains to form the complete tetrameric immunoglobulin. The transient expression of mAbs is important for conducting exploratory tests, aiming at characterizing mAbs and selecting the best candidates. Likewise, we must express recombinant proteins, such as antigens, to carry out assays.
