Abstract

Despite technological and scientific advances in bone regeneration, there is still no biomaterial with the ideal characteristics to replace large bone defects, and the association of biomaterials has been developed. The effects of bioactive glass (BG) incorporated with protein nanotubes (NT), in bone repair have been investigated; however, bone remodeling involves synergistic and controlled neoformation and resorption processes, with osteoclast participation. In this context, this study will evaluate the effect of BV-NT dissolution products on osteoclast differentiation. The RAW 264.7 cells will be cultured in either culture plastic or on bone substrates in osteoclastogenic medium conditioned with BG and BG-NT. Cell proliferation will be evaluated by MTT, gene expression of osteoclastic marker genes (tartar-resistant acid phosphatase, nuclear factor kappa B activating receptor, cathepsin K and associated receptor to osteoclast) will be evaluated by RT-PCR, and osteoclastic activity evaluated by scanning electron microscopy. Analyses of quantitative data will be conducted using appropriate statistical tests with a significance level of 5%. The results of this study may contribute to the understanding of the role of BG-NT in bone remodeling, focusing on resorptive activity, favoring the interaction between bone tissue and biomaterials
