Analysis of the protective effect of different proteins/peptides (CaneCPI-5, MaquiCPI-3, Statherin and Hemoglobin) added in orodispersible films for the prevention of initial dental erosion in vivo

Abstract

Currently, the use of proteins/peptides through acquired pellicle engineering demonstrated protection against dental erosion, using two application vehicles (solution and gel). However, there is a lack of studies with these treatments in in vivo protocols, as well as the absence of studies with another vehicle, for example, orodispersible film. Thus, the objective of this study will be to analyze different proteins/peptide (CaneCPI-5, MaquiCPI-3, Hemoglobin and Statherin), isolated or in combination through a new application vehicle (orodispersible films) for the prevention of initial dental erosion in vivo. Regarding the methodology, sixteen participants will be selected. The orodispersible films will be prepared with carboxymethylcellulose and sorbitol, using the tape casting method. The filmogenic solution will be dispersed on an automatic film spreader. Then, the study will be crossed and double-blind (8 phases): 1- Without application of the orodispersible film (negative control); 2- Untreated orodispersible film (placebo); 3- 1 mg F (positive control); 4- 0.1 mg/ml CaneCPI-5; 5- 0.5 mg/ml MaquiCPI-3; 6- 1.0 mg/mL Hemoglobin; 7- Statherin Peptide - StN15pSpS 1.88 x 10-5 M; and 8- Proteins/peptide combination. For each phase, the following experimental procedures will be performed on the maxillary and buccal central incisors: prophylaxis, initial analysis of the surface reflection intensity (SRI) using Reflectometer Optipen, treatment with or without the orodispersible film (3 X 3 cm) on the tongue (1 min), formation of acquired enamel pellicle (2 h), erosive challenge (10 µL of 1% citric acid, pH 2.5 for 10 s) and final SRI. The acid will be collected and analyzed by the Arsenazo III colorimetric method. The values obtained by the SRI will be calculated (%SRI = (SRIfinal / SRIinitial) × 100). Finally, the data will be checked for normality and homogeneity, for the use of the appropriate statistical test (parametric or not, considering p<0.05).
