Abstract

Experimental models using rodents are critical to evaluating the efficacy of bone tissue regeneration therapies. Our research group have been investigating the effect of injection of mesenchymal stem cells (MSCs) on the repair of defects in rat calvaria, with interesting results, although regeneration has not yet been achieved. Therefore, it is necessary to investigate the molecular, cellular and tissue mechanisms involved in the repair process. Therefore, the experimental mouse model is interesting because it allows for more in-depth investigations using widely available genetic models, combined with the availability of biological tools, rapid turnover and occurrence of biological processes, experimental reproducibility with less variability and reduced cost. The standardization of defects and treatments is essential for advancing these investigations. Based on these, the aim of this study is to evaluate the effect of different cell densities on bone repair in mouse calvaria. For this, 3,5 mm bone defects will be treated with injection of MSCs at concentrations of 2x106, 3x106 and 4x106, and bone formation will be evaluated by computed microtomography analysis after 04 weeks. Quantitative data will be submitted to a normality test to select the appropriate statistical test (with a significance level of 5%.)
