Abstract

Different concentrations of calcium titanate (CaTiO3) will be added to an at-home bleaching gel with 22% carbamide peroxide (CP22%). The products will be assessed concerning their bleaching effectiveness (BE), trans-enamel dentinal cytotoxicity (TC) and effects on the microstructure of enamel. For this, standardized enamel/dentin discs will be obtained, stained and bleached for 15 days (2 hours/day) with experimental gels containing 2 mg/mL, 6 mg/mL or 10 mg/mL of CaTiO3. Discs not bleached or bleached with a CP22% gel without CaTiO3 addition will be used as negative and positive controls, respectively. The total BE (n=8) will be determined by means of color measurements to be performed immediately after completing each bleaching procedure (”E00 and ”WI). To evaluate the TC, enamel/dentin discs (n=8) will be fitted into artificial pulp chambers. After completing the bleaching treatments, the extracts (culture medium + components of the bleaching gels diffused by the discs) will be collected and applied on odontoblast-like MDPC-23 cells, which will have their viability evaluated quantitatively (MTT) and qualitatively (Live/Dead). The quantification of the total H2O2 diffused by the enamel/dentin discs (n=8) will also be determined (Leuco-crystal Violet/Peroxidase). The microhardness of enamel submitted to bleaching procedures, as well as its chemical and structural characteristics (Scanning Electron Microscopy and Energy Dispersive Spectrometry) will also be analyzed (n=8). Numerical data obtained through the application of laboratory protocols will be submitted to specific statistical analysis.
