Abstract

The bone marrow tumor microenvironment plays an essential role in the progression of leukemic cells. The reciprocal interaction between neoplastic, stromal, and endothelial cells, as well as the interaction of these cells with components of the extracellular matrix and soluble factors creates an environment that drives tumor development and progression, in addition to affecting the response to therapy. Therefore, the development of in vitro methodologies that resemble the dynamics found in vivo are relevant for the study of tumor biology and for the development of new therapeutic interventions. Thus, during the fellow's internship period, we propose the development and/or improvement of methods for analyzing the bone marrow hematopoietic niche based on a 3D co-culture using a hydrolyzed collagen hemostatic sponge (gelfoam) to mimic the extracellular matrix and cells that make up the tumor microenvironment in the bone marrow, such as leukemic, mesenchymal stromal and endothelial cells. Since this model ideally imitates the pathophysiological conditions and the natural microenvironment, it becomes a fundamental platform for testing antineoplastic drugs that are already being tested at the Center for Hematology and Hemotherapy at UNICAMP. Furthermore, we aim for the scholarship holder, through working on the project, to develop new laboratory skills and apply the knowledge acquired in their projects at the institution of origin, which also seeks the development of 3D co-cultures.
