Abstract

The project "Impact of different dengue genotypes on the pathophysiology and severity of the disease: a multi-omics approach" aims to understand the changes in the dengue pathogen-host interaction, investigate whether there is any genetic factor, whether from the host or the virus, responsible for the severity of the disease, and understand if there are any proteins, peptides, or smaller molecules that may also be correlated with the severity of the infection. It is worth noting that understanding of the pathophysiology and gene variation of both the virus and the host is still scarce. In the current scenario, we have 2 hypotheses regarding the severity of the disease: the first one says that the severity is directly related to the virulence of the strain, and the second one says that the severity of the disease is related to the genetics of the host and its immune system. In this sense, it has currently been demonstrated that children and young people have been the main population affected by the severe form of the disease. This data points to a significant change in the severity of the disease. Several factors influence the effectiveness of vaccines, therefore it is important that these factors are continually monitored, so that vaccines always offer greater effectiveness. Therefore, this study, through omics tools, such as viral and human genomics, proteomics and metabolomics will contribute to obtaining fundamental data for a more robust discussion, as they will provide, in addition to understanding biomarkers related to the severity of the infection and to the predictions for severity, understanding the pathophysiology of the disease and which biological factors lead to its worsening.
