Chitosan coatings on titanium: surface characterization and microbial profile

Abstract

The progress in the characterization of implants has led to the development of surfaces that are capable of, at the same time, promoting osseointegration and prevent bacterial adhesion and proliferation, reducing the failures in the treatment. The modification on the titanium surface by the addition of chitosan contribute to minimize the adhesion of microorganisms on dental implants. The objective of this study is develop the activation of titanium surfaces with chitosan, using Layer-By-Layer (LBL) and Spin Coating techniques, and evaluate the formation and composition of oral biofilm, in situ, compared to machined titanium surfaces and the commercial surface with dual acid-etch attack. 48 pure titanium discs will be used, divided into 4 groups (n=12) according to the proposed surface treatment: C1 (control), titanium disc machined; C2 (commercial control), titanium discs with surface treatment by dual acid-etch attack; T1 (test), titanium discs treated with incorporation of chitosan by the Layer-By-Layer technique; and T2 (test), titanium disc treated with incorporation of chitosan by the Spin Coating technique. It will be realized analysis of confocal microscopy and SEM/EDS for the characterization of topography, surface properties, morphology and chemical and structural composition of the experimental and control coating surfaces, in addition, will be realized microbiological analysis by the methodology of Checkerboard DNA-DNA hybridization for characterization of microbial profile and Live/Dead cell viability assay
