Abstract

Breast cancer it's the world's most common cancer in women. This pathology can commit pregnant women, representing the pregnancy-associated breast cancer disease. The diagnosis of breast cancer during gestation it's rare but the incidence has been growing in the last years mostly because women have the option to postpone gestation. The association with breast cancer, gestation and chemotherapy, major treatment for the disease, can cause placental damages and consequently cause intrauterine growth restriction, premature birth and low birth weight. Despite importance, the responsible molecular mechanisms of systemic and placental alterations caused by breast cancer during gestation haven't been known. In this way global investigations to evaluate the communication by different organs and tissues are interessants. That way studie circulant extracellular vesicles , and more specifically this vesicles interior miRNAs, makes possible insights of the pathophysiology between tumor, placenta and fetus. Therefore the objective of this project its evaluated, by the serum of Wistars rats with Walker 256 tumors tha have or haven't been treated with chemotherapy, the presence of circulant miRNAs expression in extracellular vesicles. For this female adult Wistar rats will be allocated in comunitary boxes and the mating will be conducted by the harem method (2 female and 1 male with the same age). After pregnancy confirmation, rats will be distributed in tree experimented groups: C: wealthy; W: Walker 256 carries; WQ: Walker 256 carries that received chemotherapy treatment with doxorubicin and cyclophosphamide. Euthanase will happen in 20fh day of pregnancy and cardiac puncture will be done to obtain serum. Serum will be processed and stored -80°C until the extracellular vesicle extraction. Extraction process included phases of isolation, counting and quality control with the vesicles, in addition to Western Blotting and miRNAs expression analysis. Its important highlight that this project its part of a bigger project that has the direct participation of a post doc researcher, a experient researcher in the extracellular vesicles and all the research group that are part of the laboratory team of the Jovem Pesquisador project (2021/08931-0), in effect until 2027.
