Evaluation of intraoral levels of Fluoride in saliva, biofilm and biofilm fluid after using experimental dentifrice containing TiF4 and Chitosan

Abstract

The present work aims to compare the effect of experimental toothpaste containing TiF4 and Chitosan (1400 ppm F-) with a commercial toothpaste Elmex® caries protection (GABA, Switzerland, 1400 ppm F-, AmF) about the intraoral levels of Fluoride in saliva, biofilm and biofilm fluid, in addition to determining the pH, abrasiveness and the concentration of Total, Soluble and Ionic Fluorides. For this, 20 healthy adult volunteers will participate in four crossed phases (7 days each, 10 days washout) and double-blind, in which they will be randomly assigned to four possible treatments: 1) Experimental toothpaste containing TiF4 (1400 ppm F-) plus Chitosan 0.5% (75% deacetylation, 500 mPas, pH 4.50); 2) Experimental toothpaste containing TiF4 (1400 ppm F-, pH 4.50); 3) Elmex® caries protection commercial toothpaste (GABA, Switzerland, 1450 ppm F-, AmF, positive caries control, pH 4.55); 4) Placebo toothpaste without F (negative control). On the morning of the 7nd day of each treatment, the first samples of saliva and dental biofilm (right side of the mouth) will be collected. The collection will be performed again after 1 hour (left side of the mouth). Thus, samples will be collected 1 h and 12 h after the last use of toothpastes. Fluoride analysis of all samples (saliva, biofilm and biofilm fluoride) will be performed, and the respective concentrations tabulated in an Excel spreadsheet, and the data will be submitted to appropriate statistical analysis (parametric or non-parametric test) for comparison between treatments, considering a significance level of 5%.
