Abstract

This randomized clinical trial will avaluate the inflammatory profile and bone metabolismo in single-rooted teeth with Pulp necrosis and apical periodontitis (AP) after biomechanical preparation and the use of diferente intracanal medications. To achieve this, 45 single-rooted permanent teeth from patients with primary endodontic infection and AP will be selected. The shaping will be performed using a single reciprocating file and irrigation with 2,5% sodium hypochlorite. At the endo f the preparation, the canals will be filled for 14 days with the intracanal medications to be evaluated: calcium hydroxide [Ca(OH)2] + saline solution (n=15); Ca(OH)2 + chlorhexidine (CHX) (n=15); and Ca(OH)2 + N-acetylcysteine (NAC) (n=15). Periapical fluid samples will be collected at the end of the mechanical shaping in the first session (SF1) and after 14 days of intracanal medication (SF2) for subsequent analysis of inflammatory cytokines: IL-4, IL-17, TNF-±; matrix metalloproteinases: MMP-8, MMP-13, and bone markers: RANK-L, OPG and sclerostin. The obtained data will be subjected to normality tests, and those that do not pass the test will be transformed logarithmically and subjected to one-way ANOVA testing.
