Effect of Liraglutide on the sympathetic activity of white adipose tissue in normal and obese mice.

Abstract

Liraglutide (LIRA) is a GLP-1 (Glucagon Peptide Type I) analog drug widely used for treating Type 2 Diabetes Mellitus and obesity that results in improved glycemic control, satiety, and intense weight loss. Although there is evidence that central actions of LIRA mediate part of these effects, very little is known about the role of the Sympathetic Nervous System (SNS) in the loss of fat mass induced by the drug. The present work aims to investigate LIRA's effects on the sympathetic activity of white adipose tissue (WAT) in normal and obese mice. Through the use of electrical nerve recording techniques and quantification of norepinephrine content by HPLC, sympathetic activity will be evaluated in different WAT deposits (retroperitoneal, epididymal, and inguinal) of mice (C57/Bl6) submitted to a balanced or High-fat diet, and treated or not with LIRA acutely or chronically (Victoza®, 200¼g/kg, i.p, 2x/day), for 10 days. Pair-fed controls will be used if the treatment reduces food consumption. The results obtained may contribute to a better understanding of the mechanisms of action of LIRA in combating weight gain and complications of metabolic diseases.
