Abstract

This project aims at establishing a time-resolved cryo-EM methodology pipeline, which will include the testing and optimization of a previously developed hardware, together with data processing. This pipeline will allow to investigate macromolecular structural intermediates formed during reactions. Different biological macromolecules will be investigated, using the time-resolved approach, aiming at a wide validation of the hardware and software being developed. Among them, it will be investigated the early steps on the formation of the transcription initiation complex, by the RNA Polymerase (RNAP) and a promoter.
