Genetic variants associated with response to the treatment of people with refractory depression: a systematic review

Abstract

Introduction: Treatment-resistant depression (TRD) is a highly prevalent health problem. Although there is no consensus, it is defined as a failure to respond to the use of antidepressants, even when associated with alternatives. In this context, studies have been investigating the factors associated with treatment resistance and there are hypotheses about the influence of genetic susceptibility. Objective: To identify genetic polymorphisms associated with response to treatment in people diagnosed with TRD. Methodology: A systematic review will be conduct in the Pubmed, Scopus, Lilacs and Psyinfo databases following the guiding question: "What are the genetic variants potentially associated with the lack of response in the treatment of people living with TRD?". Will be included interventional and observational studies that evaluated the influence of genetic variants in TRD. There will be no restriction to the type of the evaluated treatment (e.g., antidepressants). Two independent researchers will conduct the selection of studies (screening and eligibility), data extraction and the analysis of the methodologic quality of the studies with the tools ROBINS-1, ROB-2 and Critical Appraisal Tools of The Joanna Briggs Institute, depending on the study design. Expected results: It is intended to identify the genetic variants associated with the response to treatment of TRD to provide evidence that can aid health decision-making and promote the safety and effectiveness of pharmacotherapy.
