IN VITRO EVALUATION OF A NEW RECOMBINANT CYSTATIN FROM ORANGE (CSINCPI-2) FOR DENTIN EROSION PROTECTION

Abstract

Recently, a new orange-derived protein, designated as CsinCPI-2, was developed. However, its potential use in the field of Dentistry, particularly regarding dentin erosion, has not yet been scientifically investigated. Therefore, this study aims to evaluate the protective effect of CsinCPI-2 and determine its optimal concentration for preventing dentin erosion in an in vitro model. For this purpose, 105 human dentin samples (4 x 4 mm) will be prepared and divided into seven groups according to the following treatments (n/group = 15): 1) Deionized water (negative control); 2) Commercial solution Elmex¿ (positive control); 3) 0.1 mg/mL of the sugarcane-derived protein (CaneCPI-5; organic positive control); 4) CsinCPI-2 at a concentration of 0.01 mg/mL; 5) CsinCPI-2 at a concentration of 0.1 mg/mL; 6) CsinCPI-2 at a concentration of 0.5 mg/mL; and 7) CsinCPI-2 at a concentration of 1.0 mg/mL. Initially, the dentin samples will be treated with the respective solutions (250 µL, 2 hours, 37°C, 250 rpm). Next, the Acquired Pellicle will be formed (250 µL, 2 hours, 37°C, 250 rpm) by the addition of unstimulated human saliva. Subsequently, the erosive challenge will be performed (1 mL, 0.65% citric acid, pH = 3.4, 1 min, 25°C, 250 rpm). These procedures will be conducted once a day for 3 days. Surface microhardness analyses and surface reflection intensity analysis will be performed (before and after the experiment), and surface changes will be calculated. The data will be analyzed for normality (Kolmogorov-Smirnov test) and homogeneity (Bartlett's test) to select the appropriate statistical test. The significance level will be set at 5%.
