Abstract

Abstract:The use of combeite as an endodontic biomaterial component becomes really promising by your osteogenic potential and your antimicrobial capacity, given the microorganism permanence is still the biggest factor of failure on endodontic treatments. This project has the main goal to enhance the antimicrobial and osteogenic properties of the endodontic cement MTA (Mineral Trioxide Aggregate) by adding fractions of combeite, which represents a bioactive phase with antimicrobial effect. The combeite will be synthesized by via Sol-Gel method, utilizing silicic acid and nitrates, follow by calcination, milling and sieving. The material will be characterized by X-ray diffraction and by energy dispersive spectroscopy (EDS), being next incorporated with MTA in different proportions of combeite (1%, 3% and 5%). Right away, the samples will be evaluated by their biological behavior through cellule culture medium using MG-63 cell line. Morphology and cell proliferation will be performed after 3 days, the cytotoxicity tests, total protein and alkaline phosphatase (ALP) after 7 days. Next in 14 days, it will be observed the mineralization of the nodules, and the calcium quantification. After that, the monotypic biofilm quantification will be carried out using standardized suspensions (107 cel/ml) of Pseudomonas aeruginosa and Staphylococcus aureus, above the different concentrations of the material after 24h of incubation. The resulting data are going to be submitted to normality test and following to adequate statistical analysis, considering the significance level of p<0,05.
