Evaluation of experimental dentifrices containing epigallocatechin 3-gallate and chitosan nanoparticles in control enamel demineralization: in situ study

Abstract

The objective of the present study will be to evaluate, in situ, the effect of experimental dentifrices containing epigallocatechin 3-gallate (EGCG) encapsulated with chitosan nanoparticles on the process of dental demineralization and the chemical composition of the biofilm. Enamel blocks from bovines (4 mm x 4 mm, n=200) will be selected based on initial surface hardness (SHi). Subsequently, twelve participants (n=12) will use palatal devices containing 4 enamel blocks, for 7 days in 4 experimental phases, with the respective dentifrices: 1) Without F/ChNP/EGCG (Negative control); 2) 1100 ppm F (1100F); 3) 1100 ppm F associated with 0.125% ChNP (1100F-ChNP) e 4) 1100 ppm F associated with 0.125% ChNP encapsulated with 0.04% EGCG (1100F-ChNP-EGCG). Cariogenic challenges will be produced with 30% sucrose solution, 6x/day. After 7 days, the final surface hardness (SHf) will be determined to calculate the percentage of loss of surface hardness (%SH), integrated loss of subsurface hardness (¿KHN), microbiological analysis of biofilm (weight and Colony Forming Units) and concentration of fluoride (F), calcium (Ca) and phosphorus (P) in biofilm. Statistical analysis will be carried out using the Sigmaplot© statistical software program for Windows version 12.0, with significance at the 5% level. (AU)
