Participation of angiotensin II upon induced-vascular injury in diabetic mice

Abstract

Diabetes mellitus is a disease characterized by several metabolic alterations, and the premature atherosclerosis is between the biggest complications. The hyperglycemia is the clinical primary demonstration in diabetes that alters the vascular metabolism in an important form. Several studies have been showing interaction between the alterations related to diabetes and the renin-angiotensin system, suggesting that the increase of his activity makes the pathologies worse when diabetes was associated to. The objective of this study is it of evaluate in which form the increase of the time of diabetes is related to the increased magnitude of vascular induced-injury. There will be used male mice C57BL/6J arranged in groups: control and diabetic, accompanied for 16, 30, 90 and 180 days. The glycemia will be evaluated weekly for the groups 16 and 30 days, and fortnightly for the chronic groups, with 90 and 180 days. The vascular injury will be induced by the implant of polyethylene catheter around the carotid and femoral arteries at the lefts side, being the right arteries like control. The arteries will be studied after 15 days of induced-vascular injury. The components of the renin-angiotensin system will be studied to connect it with the magnitude of the vascular injury due to the time of diabetes. For such, there will be done analysis of the cellular types involved with angiotensina converting enzyme, angiotensin II and angiotensina II receptor AT1, by immunohistochemistry. Depending on the obtained results, these components will be evaluated by quantitative method (Western Blotting) or direct dosage for HPLC (for angiotensina II). The obtained results will be analyzed statistically, comparing the groups for analysis of variance (ANOVA) of one or two ways, complemented by appropriated post hoc test. (AU)
