Abstract

The Bradysia hygida (Diptera, Sciaridae) DNA puff C4 BhC4-1 gene is amplified and expressed in a developmentally regulated manner in the salivary gland at the time DNA puff C4 is formed. Since it is not possible to perform functional assays in Sciaridae, the BhC4-1 regulation is studied in Drosophila, a system in which the pattern of BhC4-1 expression is largely conserved. In transgenesis the onset of BhC4-1 expression is verified in prepupal salivary glands and in the ring gland of late embryos. The CIs-regulatory elements which control gene expression in the salivary gland are separable from those which regulate gene expression in the ring gland. Very little is known about ring gland specific transcription factors. In this sense, this project aims to identify trans-activating factors which regulate BhC4-1 gene expression in the ring gland employing the yeast one-hybrid system, a tool which has been widely employed to identify transcription factors. The obtained results will contribute for the understanding of the regulatory mechanisms which control developmentally regulated and tissue-specific gene expression. (AU)
