Abstract

Parkinson´s disease (PD) is characterized by motor alterations resulting from nigrostriatal dopaminergic cells degeneration, which modifies the amount of dopamine in striatum, unbalancing the motor circuit responsible for the normal movements. Genetic predisposition, aging and environmental toxins contribute to PD etiopathogenesis. Apolipoprotein E, that is found in high concentrations in the brain and is codified by three distinct alleles named APOE*2, APOE*3 and APOE*4, has been considered a risk factor for PD. In this context the APOE*4 allele seems to be associated to neurodegeneration in PD. This study aims to identify the association of apo E genetic polymorphisms with familial and sporadic PD and evaluate its influence in the age of PD onset and in the pacient's lipidic profile. One hundred and fifty patients with PD and 150 individuals without PD (controls) will be studied. The apo E polimorphisms will be analysed by conventional polymerase chain reaction (PCR), in 1,5% agarose gel and stained with ethidium bromide. The post PCR product will be submitted to RFLP (restriction fragment length polymorphism) with restriction enzyme Hha I. The statistical analysis will consist in Fisher test or chi-square and t-test or Mann-Whitney and analysis of variance, with level of significance for P<0.05.
