Abstract

The bone is a dynamic tissue under constant turn-over regulated by differentiation and activity of cells such as osteoblasts and osteoclasts, modulated by a wide variety of hormones, inflammatory mediators and growth factors. These mediators are found in periodontium (gingival and bone), blood, crevicular fluid and saliva, being the last an important oral fluid easy to collect and rich in variety of substances, whose analysis can be useful to correlate with the oral health status.In this way, the aim of this project is to investigate whether the saliva from healthy individuals or patients with periodontal disease may interfere with the mechanisms of bone resorption, either through a direct interaction with osteoclasts or indirectly through osteoblasts, and if these events are related to the clinical status of patients with periodontal disease. In this study, we will analyze the effects of saliva on the differentiation and activity (bone resorption) of of osteoclast precursors, using pure osteoclast cell cultures obtained from human peripheral blood monocytes, co-cultures with human osteoblasts and the intracellular mechanisms involved, through the analysis of protein and gene expression of some mediators (such as osteoprotegerin, AkT phosphorylation, Traff6 and NF-kB).
