Evalution of C. albicans cariogenicity isolated from pedriatric patients with sickle cell disease.

Abstract

Due to the increased risk for infections, patients with sickle cell disease need to take the antibiotic for a long period of time. The aim of this study is to evaluate de acidogenicity and extracellular polysaccharides production of C. albicans biofilms previously isolated from 25 children with sickle cell disease aged between 4 and 6 years old. For comparisons, 25 samples from healthy patients from the same gender and age ± 2 years will be included. The isolates will be stored at -20°C in BHI broth with glycerol 10% until analysis. Species identified as C. albicans will be analyzed regarding lactic acid and extracellular polysaccharides production in biofilm. The biofilms will grow for five days in microtiter plates. The medium will be refreshed on a daily basis. The lactic acid production will be analyzed by the enzymatic method. The extracellular polysaccharides present on dried biofilm and will be extracted with NaOH 1.0 mol L-1. The carbohydrates concentration will be colorimetrically determined. The data will be analysed with the software GraphPad Prism 3.02. The significance level will be set at 5%.
