Abstract This study aims to optimize the conditions of amplification of microsatellite markers and to assess the genetic diversity of 22 accessions (landraces and commercial varieties) of yams (Dioscorea trifida) collected in traditional agricultural systems of Vale do Ribeira (SP), in the municipalities of Cananéia and Iguape, as well as in traditional communities and markets in the Amazon and indian villages in the State of Mato Grosso. For the DNA extration, the method of Elias et al. (2004) will be tested, based in the CTAB extration buffer, with modifications. Also, the conditions of amplification and revelation of 11 primers will be tested for the assessment of the 22 accessions of this species. Being a polyploid species, data will be analysed as presence or absence (+/-) of bands and thus submitted to the variance analysis (AMOVA) using the Arlequin program. Also, a principal component analyses and a cluster analyses will be carried out, using the coefficient of Jaccard and UPGMA method, with the softwares SAS for windows and NTSYS-PC. | |
