Defining the role of cementocytes in dental cementum homeostasis

Abstract

Cementocytes and osteocytes share morphological features, and both have very similar phenotype/genotype. Different from osteocytes, cementocytes biological role is still undefined. Thus, we hypothesized that similar to osteocytes in bone tissue, cementocytes play an essential role in periodontal tissues biology. Therefore, we determined the potential participation of cementocytes in the process of neoformation of dental cementum in control animals and knockouts for the phosphate regulating endopeptidase homolog gene X-linked (PHEX), using the supereruption model (EIA). We also evaluated ultrastructural changes in cementocytes and cellular cementum proteome in response to orthodontic movement (OTM) in rats. Tissue and cellular response to EIA were analyzed at 6 and/or 21 days post-procedure (dpp) by a combination of laser scanning confocal microscopy (CLSM), histomorphometry, computed micro-tomography (Micro-CT) and transmission electron microscopy (MET). Cementocytes response against OTM (0.5N) was determined using histological evaluation (H&E), MET, and Micro-CT. Besides, changes in cellular cementum proteome were examined using liquid chromatography coupled to tandem mass spectrometry (LC-MS / MS), and target proteins were validated by immunohistochemistry (IHC). For the first time, we present evidence on the participation of cementocytes in the control of dental cementum homeostasis, both during the process of neoformation and in response to mechanical stimuli (OTM). These findings improve understanding of cementum biology and provide additional knowledge towards controlling formation and regeneration processes of dental cementum in the future (AU)
