Use of bisphosphonates and pentoxifylline on induced periodontitis bone modulation. Study in rats

Abstract

The aim of this study was to evaluate the process of periodontal bone repair when using pentoxifylline (50 mg/kg/day) on the prevention and/or treatment of induced periodontal disease. This project was separated in 2 studies: Study 1: to evaluate if pentoxifylline interfere on the progression of induced periodontal disease. Animals were divided in: Control Group – saline (C); Pentoxifylline Group (PTX) and Bisphosphonate Group (BIS). The animals received the medicines for 60 days; then periodontal disease was induced by ligatures on the second molar. In the moment the medicines administration was interrupted. After 15 days the animals were euthanized. The results demonstrated in the microtomographic analysis a similar result between pentoxifylline and control, in the histometric analysis less bone loss in the PTX group in the interproximal region was found and stereometric analysis showed a greater degree of inflammation in the group that received pentoxifylline. it was concluded that pentoxifylline had no effect on bone loss. Study 2: to evaluate if pentoxifylline interfere on the treatment of induced periodontal disease. Animals were divided in: Control Group – saline (C); Pentoxifylline Group (PTX) and Bisphosphonate Group (BIS). By the time the ligatures were made the animals started receiving the medicines until the moment they were euthanized. After 15 days the ligatures were removed. At this moments half of the animals received scaling and root planning with hand instruments (Control – CR, Pentoxifylline Group- PTX-R, and Bisphosphonate Group - BIS-R) and the other half of each group didn’t receive any treatment (Control – CR, Pentoxifylline Group – PTX, and Bisphosphonate Group - BIS). After the periods 7, 15 and 30 days the animals were be euthanized. The animals in the groups that received pentoxifylline had a higher percentage of blood vessels, less of connective tissue in the stereometric analysis, and a higher degree of inflammation in the BISR group. Histometric and microtomographic analyzes showed a less satisfactory result in the initial periods in the groups that received scaling and root planning, which after the period of 15 days started to have less bone loss. It was possible to conclude that the PTX and PTXR groups had a positive result in bone formation. (AU)
