Experimental study of the participation of the renin-angiotensin-aldosterone system in cardiac hemodynamic and morphofunctional adaptations caused by ovarian failure - effect of aerobic exercise training

Abstract

Deprivation of ovarian hormones in Wistar rats promotes increased expression of cardiac fibrosis, which can result in incorrect damage. In turn, aerobic exercise prevents this increase. The mechanisms involved in this process are still uncertain. Our hypothesis is that it is due to the increased expression of mitochondrial matrix proteins. The objective was to investigate whether the increase in cardiac fibrosis resulting from the deprivation of ovarian hormones is related to the increase in MMP-2 and calpain-1, and whether aerobic physical exercise modulates the expression of these proteins. The methodology used was Wistar at 12 weeks of age (N = 40) were divided into two groups: ovariectomized (OVX) and undergoing sham surgery (SHAM). Half of each group underwent aerobic physical training for 12 weeks. In the 24th week of life, the animals were found for the analysis of cardiac morphology and functionality through two-dimensional echocardiography; histological analysis for quantification of cardiac fibrosis; and quantification of Calpain-1 and MMP-2 expression. The main results were that the OVX trained groups had lower ejection fraction and cardiac index values when compared to the SHAM trained group. About Capain1 / GAPDH values, the OVX group obtained a higher value when compared to the SHAM and trained SHAM groups. While the trained OVX group had a lower value when compared to the OVX. We conclude that ovarian hormones influence the expression of calpain 1 and aerobic physical training modulates the activity and expression of this protein. (AU)
