In this project, our group identified 895 differentially expressed long non-coding RNAs (lncRNAs) associated with stemness in pancreatic ductal adenocarcinoma (PDAC) by applying the mRNA stemness index (mRNAsi) to RNA-seq data from TCGA. Based on expression levels, fold changes, genomic annotation, and biological relevance, nine intergenic lncRNAs were selected for further validation. My contribution focuses on validating the differential expression of these lncRNAs in PDAC cancer stem cell models in vitro using tumorsphere cultures and RT-qPCR analysis. As a preliminary result, I demonstrated that lncRNA ENSG00000260912 is significantly upregulated in tumorspheres derived from AsPC-1, PANC-1, and MIA PaCa-2 cell lines, indicating its association with cancer stem cell enrichment. This work may contribute to the identification of novel biomarkers and potential therapeutic targets in PDAC. (Source: Lattes Curriculum)
