Effect of bergamot leaf extract (Citrus bergamia) on metabolic, lipid, and oxidative imbalance in the liver of obese rats

Abstract

Obesity is a global health crisis associated with comorbidities such as metabolic dysfunction and metabolicassociated steatotic liver disease (MASLD). Oxidative stress and inflammation are hallmarks of obesity's pathophysiology. Citrus bergamia leaf extract (BLE), rich in antioxidants, has shown potential in reducing oxidative stress and improving lipid profiles. Thus, the study evaluated the effects of BLE on oxidative stress, fatty acid profile, and metabolites in the liver of obese rats. Wistar rats were fed a control diet (C) or a high sugar-fat diet (HSF) with 25 % sucrose in water for 20 weeks. After this period, they were redistributed into BLE-treated and vehicle-treated groups (n = 10/group). BLE was administered by gavage at 50 mg/kg for 10 weeks. Nutritional profiles, plasma and liver biochemistry, histology, oxidative stress markers, antioxidant enzymes, metabolomics, and fatty acid profiles were analyzed. Data were analyzed by two-way ANOVA or Kruskal-Wallis tests (p < 0.05). The obese group exhibited an increased adiposity index, plasma and hepatic triglycerides, plasma and hepatic cholesterol, glucose, insulin resistance, a higher percentage of lipid droplets in the liver, and liver oxidative stress. BLE-treated obese rats showed reduced body weight, adiposity, and hepatic cholesterol and improved HDL, oxidative stress markers, and antioxidant enzyme activities. Metabolomics revealed that deregulated metabolites in the liver of obese mice that BLE normalized were associated with liver dysfunction. BLE treatment improved the fatty acid profile of obese rats, which suggested inflammatory and oxidative metabolism. These findings indicate that BLE attenuates obesity-related liver damage, improves oxidative stress, and modulates lipid and fatty acid metabolism. (AU)
