Antibodies IgY production against yeast in sugar cane silage

Abstract

The aim is to produce and evaluate the effect of polyclonal antibodies against yeast in sugar cane silage. Three hundred hens White Leghorn race with 25-weeks-old divided into groups of 75 individuals will be immunized, one control immunized only with adjuvant and four others groups will be immunized with strains a) Candida, Endomycopsis, Hansenula e Pichia (use acid latic), Torulopsis (use sugar) e primitive yeasts. Three intervals between the immunizations (14, 28 and 42 days) will be evaluated, will be carried 6, 3 and 2 immunizations, respectively, during a period of 84 days. The yolk will be delipidation and freeze-dried. The ELISA and microbiological assays will be performed to verify the concentration and antifungal effect of the IgY. Fourthy experimental silos will be used to evaluate eight sugar cane IAC 86-2480 silage with or without the IgY and with or without L. buchenri (according factories recommendation). The experimental silos will be exposed to air after at least 100 days of fermentation, the silages will be placed in a polystyrene box, and transferred to a climatic chamber kept at 25 ± 1oC to determine the temperature and pH values. Thermometers will be inserted 10 cm into the silage mass, the temperature will be recorded at every 5 min and pH values twice daily lasting 5 days. The variables measured will be dry matter losses, organics acids concentration, chemical composition, yeast and fungi population and aerobic stability. The aerobic stability will be defined as the time in min to elevate the temperature by 2ºC in relation to environmental temperature. Beside this will be determined maximum temperature reached by the mass in ºC, hours to reach maximum temperature and heating rate measured. The data will be analyze according to a repeated measures model in a complete randomized design with eight treatment and five replications, in factorial arrangement 2 x 2. The data will be submitted to the analysis of variance by the PROC GLM of the SAS and the Tukey test at the significance level of 5%. (AU)
