Abstract

Anthracosis is a lung injury characterized by pigmentation of the cells by endocyte carbon particles and may cause diffuse pulmonary fibrosis. It is believed that this disease is a result of damage to different areas of the alveolar epithelium, resulting from dysregulation of cellular homeostasis, followed by aberrant scar healing and inadequate repair of epithelial damage, and one hypothesis for the progression of this disease is deregulation of autophagy. One way to study the effects of certain cells on particle exposure is to co-culture two types of cell (epithelial and inflammatory) to assess relevant aspects of this interaction. This project aims to study autophagic flow, cytokine production and expression of CYP450 in co-culture of A549 and M1 or M2 cells when exposed to particulate material collected from atmospheric pollution (PM2.5) and anthracosis collected from the corpse of the USP Capital Surveillance Service (SVO), relating the outcomes to the anthracosis pathophysiology.
