Abstract

Detection, monitoring and quantification of molecular processes in simple systems, such as biochemical reactions in isolated systems, or complex ones, such as those occurring within cells or supramolecular systems, occurs by contrasting between what is wanted and what is not wanted to see. In other words, by increasing the signal to noise ratio. In this scenario, fluorescence-based techniques stand out because they are the most used methods in chemical and biomedical field. Indeed, it would not be possible to imagine the development in areas as biochemistry, immunology, microbiology, cell biology, molecular biology etc, without the use of fluorescent markers capable of elucidating the expression of proteins, cytokines, metabolites, reactive oxygen and nitrogen species, release or uptake. ions, changes in intracellular pH, membrane potential, signaling pathways, transcription factors etc? This project fits in this field, since part of it is focused on the development and application of fluorescent probes for the following purposes: synthesis and study of fluorescent aminoquinolines for selective determination of enzymatic activity of the enzymes myeloperoxidase and eosinophil peroxidase; synthesis and study of fluorescent probe for DNA study via exciplex formation; energy transfer study (FRET) for elucidating interaction of fluorescent probes with albumin; solvatochromic studies with the probes 4-(Dicyanomethylene)-2-methyl-6-(4-dimethylaminostyryl)-4H-pyran and 4-dimethylamino-²-nitrostyrene and application for determination of dielectric constant inside protein. The project also aims the synthesis and development of probes susceptible to induction of circular dichroism signal. In this case we aim to develop a method to monitor the displacement of the probes between two proteins as a function of time and to apply to the characterization study of amyloid aggregates. Also, in this line, the circular dichroism signal induced in rose Bengal dye will be used to study its movement in the protein. All the studies mentioned above will have their theoretical counterpart by ab initio methods which will seek to understand the phenomena at molecular level, simulations of absorption, emission and circular dichroism spectra of free and complexed probes to macromolecules.
