In vivo analysis of local and systemic migration of ions from ProRoot MTA and tricalcium silicate

Abstract

Reparative cements such as the MTA are widely used in endodontics in procedures that include sealing of perforations, pulp capping, apical surgeries, and apexifications. The composition of such cements includes the presence of bismuth oxide, as a radiopacifying agent. Bismuth ions are very reactive and interact chemically with dental structures, resulting in the deposition of a black precipitate that causes dental discoloration. Recent studies have shown that there is a migration of ions from such materials into adjacent tissues. The objective of the study is to evaluate the migration of bismuth ions from the ProRoot MTA cement locally and systemically after contact with connective tissues and bone. A total of 64 animals will be used for the evaluation of ProRoot MTA (TCS-BiO) cements (n = 20), containing bismuth, as compared to tricalcium silicate cement (TCS), without bismuth (n = 20). Two samples of each cement (4 mm in diameter x 2 mm height) will be implanted in the subcutaneous tissue or femur, with each animal receiving two implants of the same cement. As a positive control, 20% bismuth oxide associated with hydroxiapatite (HAp-BiO) will be implanted (n=20). As a negative control (n = 4), the animals will not receive any type of implant. After the period of 30 days, half of the animals will be sacrificed and after 180 days the others. After euthanasia, the local tissues, total volume of blood and organs: brain, liver and kidneys will be collected. SEM and elemental maps of local tissues will be performed. For systemic analysis, inductively coupled plasma-mass spectrometry for the presence of bismuth and silicon ions will be used. The implanted cements will be submitted to XRD analysis to determine the chemical interaction with tissues. The results will be submitted to appropriate statistical analysis with significance level of 5%. (AU)
